El descubrimiento y conquista de la América española fue hecha por aventureros codiciosos y suertudos, como el mismísimo Colón en 1492. Venían comprometidos a sumar nuevos territorios y provincias étnicas a los dominios del Rey, al tiempo que debían realizar «la cosecha de las almas» con la conversión de los infieles a la doctrina cristiana.
Fue bajo este esquema que casi medio siglo después las huestes de Ortún Velasco (natural de Cuellar en Segovia} y Pedro de Orsúa (natural de Pamplona en Navarra} llegaron y conquistaron los pueblos chítareros que habitaban estos intrincados valles andinos, fundaron la ciudad de Pamplona de Indias en 1549, repartieron y encomendaron sus comunidades indígenas e iniciaron la hispanización de toda una gran región, incluyendo los exuberantes y fértiles valles de Cúcuta.
En algunos casos hubo enfrentamientos con fuertes guazábaras a los nativos, como ocurrió en el «Valle de Los Locos» (Labateca y Toledo} donde los indios, por no acatar el requerimiento del español y resistirse al invasor, fueron aperreados y algunos de ellos muertos. En la mayoría de los pueblos chitareros, como los cúcutas, la conquista fue pacífica.
